摘    要:本文以高效液相色谱(HPLC)法研究了广谱抗蠕虫药丙硫苯咪唑在兔体内的动力学特征。检测了兔以20mg/kg 单剂量 ig 后,血浆及心、肝、肾、肌肉等组织中丙硫苯咪唑、代谢物亚砜及砜、药物总浓度的 Tmax,Cmax,t(1/2),AUC 等药动学参数;分析了血浆及各组织中丙硫苯咪唑及其代谢物的相对比例;对血药总浓度与各组织中药物总浓度,组织药物总浓度与血浆药物总浓度的比值,对相应给药后的时间进行了回归分析。

Abstract:Pharmacokinetic processes of albendazole in rabbits were studied by HPLC proce-dure;regression analysis methods were used for studying the dynamic relationship ofdrug concentration between plasma and various tissues;and the relative percentage of thethree drugs in plasma and various tissues were analysed.Pharmacokinetic results showed that rabbits orally given albendazole as a suspensionat a dose rate of 20 mg/kg,the parent drug was not dete ctable in plasma at any time,but its two major metabolites,sulfoxide and sulfone,were measured in larger quantities,and the total concentration profiles could be described by an one-compartment openmodel with first-order absorption.Albendazole and its two major metabolites weredetectable in heart,liver,kidney and muscle,but the percentage is different in various tis-sues.The primary parameters of total concentration in plasma,heart,liver,kidney andmuscle were as follows:Tmax:4.37,3.20,8.22,4.45,4.87(h)Cmax:4.63(μg/mL),7.44,16.32,12.56,5.03(μg/g)t(1/2):10.36,7.04,8.56,11.34,7.04(h)AUC:92,76(μg.h/mL),103.50,439.68,263.31,82.57(μg.h/g)The regression analyses of the total concentration in plasma and various tissues weremade.The results indicated that the relationship of the drug concentration betweenplasma and various tissues was highly remarkable.
