摘    要:目的]探索出玉米高频诱导具有胚性的愈伤组织和植株再生的条件。方法]以玉米自交系幼胚为外植体,研究了不同2,4-D浓度对愈伤组织诱导、6-BA浓度对试管苗分化以及IBA浓度对试管苗生根的影响。结果]2,4-D对玉米胚性率诱导有明显影响,其最佳诱导培养基为N6+2,4-D 2 mg/L+水解酪蛋白(CH)500 mg/L+脯氨酸(pro)500 mg/L+AgNO310 mg/L;试管苗分化适宜的培养基是N6+6-BA 0.5 mg/L+水解酪蛋白500 mg/L;试管苗生根的最佳培养基为1/2 MS+IBA 0.5 mg/L。结论]该研究可为玉米抗病基因的遗传转化和优良抗病玉米品种的培育奠定基础。

Establishment of Plant Regeneration System from Immature Embryos of Maize (Zea mays L.) Inbred Lines

Abstract:Objective] The aim was to explore the conditions of high frequency induction of embryonic callus and plant regeneration of maize.Method] Immature embryos of maize inbred lines were used as explants to study the effect of different 2,4-D concentrations on the induction of callus,the effect of different 6-BA concentrations on the differentiation of test-tube plantlet,as well as the effect of different IBA concentrations on the rooting of test-tube plantlet.Result] 2,4-D showed obvious effect on the induction of inducement rate of maize,and the optimum induction medium was:N6 + 2 mg/L of 2,4-D + 500 mg/L of CH + 500 mg/L of Pro +10 mg/L of AgNO3;the optimum differentiation medium was:N6 + 0.5 mg/L of BA + 500 mg/L of Pro;the optimum for the rooting of test-tube plantlet was 1/2 MS + 0.5 mg/L of IBA.Conclusion] The study had provided basis for the genetic transformation of maize resistance gene and the cultivation of maize resistance varieties.
