反义LeEIL2基因表达载体的构建及在番茄中的转化

摘    要:为了研究LeEIL2基因在番茄果实成熟过程中的作用 ,将全长的番茄LeEIL2基因以反义方向构建到植物表达载体 pBI1 2 1上 ,经过酶切鉴定正确后 ,将该载体转入农杆菌EHA1 0 5中 ,通过农杆菌介导法转化番茄 ,经组织培养和抗性筛选获得再生植株 ,PCR检测再生植株 ,初步鉴定为阳性植株 ,为获得转反义LeEIL2番茄果实打下基础。

Abstract:In order to investigate the role of LeEIL2 in tomato fruit ripening,the full-length LeEIL2 gene was inserted in reverse orientation into expression vector pBI121,and subsequently confirmed with restriction enzyme analysis. The antisense expression vector pBI121-LeEIL2 was mobilized into Agrobacterium tumefacious EHA105 by chemical-based DNA direct transformation. Transgenic tomato was developed using Agrobacterium-mediated transformation with kanamycin resistance selection. The presence of antisense LeEIL2 in transformants were confirmed by PCR and transgenic tomato fruit will be obtained later.
