苜蓿高效再生体系的建立

摘    要:以黑龙江主栽苜蓿品种肇东苜蓿、敖汉苜蓿、公农1号苜蓿、公农2号苜蓿为受体材料,分别研究了外植体类型对不定芽分化的影响,基因型、PGRs的不同配比对子叶节不定芽分化的影响,并确定了芽分化、芽伸长及再生植株生根时的最佳培养基。结果表明,当BA浓度为1.0mg/L时最适于子叶节的分化;当BA浓度为0.5mg/L时植株生长最佳。芽分化培养基为:MS+1.0mg/LBA,30g/L蔗糖,0.8%琼脂,pH5.8;芽伸长培养基为:MS+0.5mg/LBA,30g/L蔗糖,0.8%琼脂,pH5.8;再生植株生根培养基为:MS+1.5mg/LIBA,15g/L蔗糖,0.8%琼脂,pH5.8。

Abstract:The effects of different explant on buds differentiation,PGRs on adventitious buds regeneration of cotyledonary nodes and factors on regenerated plant of "Zhaodong","Aohan","Gongnong No.1" and "Gongnong No.2" were studies.The result indicated:BA 1.0mg/L was the best;When BA was 0.5mg/L,the plant developed in good state.The optimal medium for root generation was MS 1.5mg/L IBA,15g/L Sucrose,0.8%Agar,pH5.8.
