家蚕幼虫正常个体与眠性变化个体大眠蜕皮前后血液蛋白质差异表达分析

摘    要:家蚕眠性既受染色体上主基因的控制,又受其它伴性成熟基因的调控,还受环境因素的影响。为了探讨环境因素改变家蚕眠性的分子机制,以四眠蚕品种898黄绿、D92黄绿和三眠蚕品种三眠A为材料,通过催青期或小蚕期温度、湿度、营养等因素的调控诱导眠性变化,利用双向电泳和基质辅助激光解析电离飞行时间质谱(MALDI-TOF-TOF)技术,对这些家蚕正常个体和眠性变化个体大眠蜕皮前后的血液蛋白质表达差异进行分析鉴定。结果共获得转铁蛋白、胰凝乳蛋白酶抑制剂SCI-I、多聚腺苷酸结合蛋白2、27 kD糖蛋白前体、核糖体蛋白L20和线粒体核糖体蛋白L2等13个上调或下调的可能与家蚕眠性相关的差异表达蛋白,为进一步从蛋白质水平深入了解家蚕眠性变化的机制奠定了基础。

Abstract:Moltinism of the silkworm(Bombyx mori)is not only controlled by major genes on its chromosomes but also regulated by genes associated with sex maturity and influenced by environmental factors as well.In order to explore the molecular mechanism of silkworm moltinism alteration induced by environmental factors,two silkworm tetramolter varieties,898 huanglü and D92 huanglü,and one trimolter variety,Sanmian A,were used as the materials to induce moltinism alternation through controlling the factors including temperature,humidity and nutrient supply in egg incubation stage or during young larva stage.Through two-dimensional electrophoresis and matrix-assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-TOF),the differential expression of hemolymph proteins between the normal and moltinism altered larvae of the silkworm before and after the last molting were analyzed.As a result,totally 13 proteins,including transferrin,chymotrypsin inhibitor SCI-I,polyadenylate binding protein 2,27 kD glycoprotein precursor,ribosomal protein L20 and mitochondrial ribosomal protein L2,were found to be up-or downregulated,showing that they are involved in moltinism alteration of silkworm.The obtained data comprise a good basis for further elucidation on the mechanism of silkworm moltinism alteration at protein level.
