珍贵树种山白兰的组织培养研究

摘    要:目的]研究山白兰组织培养快速繁殖。方法]以山白兰盆栽苗的带芽茎段为外植体,MS为基本培养基,通过添加不同种类及浓度的植物生长调节剂,对山白兰进行了组织培养试验。结果]MS+6-BA 0.5 mg/L+NAA 0.1 mg/L为最适宜的芽诱导培养基,芽的诱导率达到88.9%;MS+6-BA 0.3 mg/L+NAA 0.05 mg/L+KT 1.0 mg/L为最适宜的增殖培养基,芽的增殖系数可达到2.6;生根诱导难度较大,在1/2 MS+NAA 2.0 mg/L上生根率达到54.5%。结论]结果为山白兰的进一步开发和应用,丰富广西林木种质资源提供参考。

Study on Tissue Culture of Rare Species Paramichelia baillonii

Abstract:Objective] The aim was to research the rapid reproduction of the tissue culture in Paramichelia baillonii(DC) Hu.Method] Taking the stems with buds from the potted seedlings of P.baillonii as the explants and MS medium as the basic medium,the tissue culture test of P.baillonii was carried out through the addition of the plant growth regulators with different types and concn.in the medium.Result] The best but induction medium was MS+0.5mg/L6-BA+0.1mg/LNAA,with the but induction rate of 88.9%;the optimum medium for proliferation was MS+0.3mg/L 6-BA +0.05mg/L NAA +1.0mg/L KT,with the bud proliferation coefficient of 2.6;the rooting induction had a greater difficulty and the medium of 1/2MS+2.0mg/L NAA could get the rooting rate of 54.5%.Conclusion] The study provided the reference for the further development and application of P.baillonii and the abundance of Guangxi forest tree germplasm resources.
