摘    要:目的]建立高效液相色谱法(HPLC)测定黄芩毛状根中熊果苷及残余底物对苯二酚含量的方法。方法]采用乙醇回流法提取加底物转化后的毛状根,并采用HPIC法测定黄芩毛状根中熊果苷及残余底物对苯二酚的含量。色谱条件:色谱柱为Symmetry-C18(250苷及对苯二酚峰型和分离效果良好,熊果苷标准曲线线性范围为0.100~0.200 mg/ml(R=0.999 1),精密度RSD为1.52%,回收率为98.6%~100.3%。样品中熊果苷的含量为45.5 mg/g;以外标一点法测得毛状根中残留对苯二酚的含量为0.22 mg/g。结论]该方法准确可靠,可用于黄芩毛状根转化产物熊果苷及残余底物对苯二酚的含量测定。

Study on the Determination of Scutellaria baicalensis Hairy Roots Converted Products-Arbutin Using HPLC Method

Abstract:Objective] The research aimed to build high-performance liquid chromatography(HPLC) determination method to determine arbutin in Scutellaria baicalensis hairy roots and residual substrate hydroquinone content.Method] Using ethanol refluxing method,extract hairy roots from converted substrates.And using HPIC method,determine arbutin and residual substrate hydroquinone content in Scutellaria baicalensis hairy roots.The chromatographic conditions were:chromatographic column:Symmetry-C18(250 mm×4.6 mm,5 μm),mobile phase was Absolute methanol-water(15∶85,V/V),flow rate of 0.8 ml/min,detection wavelength was 287 nm.Result] Under the mobile phase conditions,arbutin and hydroquinone has a good peak shape and separation effect.The standard linear range of arbutin was 0.100-0.200 mg/ml(R=0.9991),precision RSD was 1.52%,recovery rate was 98.6%-100.3%.Conclusion] This method could accurately measure the content of Scutellaria baicalensis hairy roots transformation arbutin and residual substrate hydroquinone content.
