摘    要:对会龟总科(Scarabaeoidea)6种金龟子线粒体细胞色素氧化酶Ⅰ(COI)基因部分序列进行了PCR扩增和序列测定,并从GenBank中检索4个种的COI基因序列,比较其同源性并获得了长度为624bpCOI片段.10个种的COI序列分析结果表明:该基因序列共有440个变异位点,192个简约信息位点.表现出明显的A+T偏倚(63.6%).转换(TV)和颠换(TS)在科内、科间存在明显差异.科内种问TV+TS小,各科不同种与外群之问TV+TS较大;TV/TS则呈现相反的趋势.以黑蜣科(Passalidae)的具角黑艳甲(Odontotaenius disjuncttts)作为外群,采用P-distance法重建了10种金龟子邻接树(N-J)和最小进化树(ME)拓扑结构,结果表明:丽金龟属内科内、鳃金龟科内的不同种分别优先聚集,再与其他科相聚集,显示科内、属内的种间有较近的亲缘关系,说明COI基因序列可用来研究属内或科内种群的系统发育问题.

Abstract:The mtDNA-COI gene fragments of 6 Scarabeoidea species were amplified and sequenced by PCR and the COI partial sequences of 4 relative species were retrieved from GenBank. The homologus sequences were compared and obtained partial nucleotide sequence of COI (624bp). The analysis results of 10 sequences of COI gene showed that there were 440 variable sites and 192 parsimony-informative sites in the 624 nucleotides of the data. Scarabaeoidea exhibited a 63.6% Arginine and Thymine bias. The distinct variation rate of transition and transversion among different species of intra-family and inter-family in Scarabaeoidea was determined. The numbers of TV+TS of COI sequences among different species in the same family were higher than those in the different families. The ratio of TV and TS showed the opposite trend. The topological trees of Neighbour Joining and Minimum Evolution constructed by P-distance method using Passalidae Odontotaenius disjunctus as the outgroup revealed that the species in Rutelidae and Melolonthidae were grouped together first, and showed close relationship. They were further grouped together with the species of other families. The analysis results of COI gene sequences and phylogenic threes showed that there were close relationships among species of the genus or the family. The phylogenetic relationships of the species in genera or family could be well resolved with COI gene sequences.
