摘    要:为了研究不同碳氮比(C/N)对杨梅枝条发酵堆肥过程中养分和腐殖质组分的影响,探究C/N对堆肥发酵过程中细菌群落结构变化的影响。本研究设置三个C/N梯度分别为30:1,60:1,90:1进行杨梅枝条堆肥发酵实验。结果表明:全氮(TN)、全磷(TP)、全钾(TK)的含量随C/N增加而减少,有机质(OM)和胡敏素(HM)的含量随C/N增加而增加。TN、TK、胡敏酸(HA)和富里酸(FA)的含量随发酵时间而明显增加,OM和HM的含量随发酵时间而明显减少(p<0.05);C/N与OM和TN呈极显著相关(p<0.01)。对堆体细菌群落进行16SrRNA测序分析研究发现,厚壁菌门和放线菌门的相对丰度随C/N增加而增加,变形菌门和拟杆菌门的相对丰度随C/N增加而减少;在高温阶段厚壁菌门和放线菌门的相对丰度明显高于其他阶段。通过Alpha多样性的Chaol、Faith’s PD、Pielou’s evenness、Shannon和Simpson等指标分析发现,不同C/N处理的细菌群落丰富度和多样性存在明显的差异。非度量多维度和典型对应分析表明,C/N对枝条发酵堆体细菌群落结构起着决定性的作用;TN、OM和温度也对枝条发酵堆体细菌群落结构有一定的影响。Mantel测试进一步表明,堆体细菌群落结构与C/N(r=0.321,p=0.001)、TN(r=0.265,p=0.003)和OM(r=0.278,p=0.002)之间存在显著的相关性。这些结果表明C/N对杨梅枝条堆肥养分含量和堆体细菌群落分布变化的起着关键性的作用。

Abstract:In order to study the effects of different C/N ratios on the nutrient and humus components in the process of fermentation and composting of Myrica Rubra branches, and to explore the effects of C/N on the changes of bacterial community structure in the process of compost. In this study, three C/N gradients of 30:1, 60:1, and 90:1 were set for the experiment of Myrica Rubra branch compost fermentation. The results showed that the contents of total nitrogen (TN), total phosphorus (TP) and total potassium (TK) decreased with the increase of C/N, while the contents of organic matter (OM) and humin (HM) increased with the increase of C/N. The contents of TN, TK, humic acid (HA) and fulvic acid (FA) increased significantly with the fermentation time, while the contents of OM and HM decreased significantly with the fermentation time (p<0.05); C/N was significantly correlated with OM and TN (p<0.01). The 16S rRNA sequencing analysis of the bacterial community showed that the relative abundance of Firmicutes and Actinobacteria decreased with the increase of C/N, while the relative abundance of Proteobacteria and Bacteroides increased with the increase of C/N; In the high temperature stage, the relative abundance of Firmicutes and Actinobacteria was significantly higher than that of other stages. Through the analysis of indicators such as Chaol, Faith''s PD, Pielou''s evenness, Shannon and Simpson of alpha diversity, it was found that the richness and diversity of bacterial communities in different C/N treatments were significantly different. The Non-metric Multi-Dimensionals and Canonical Correspondence Analysis showed that C/N played a decisive role in the bacterial community structure of the fermentation pile; TN, OM and temperature also have certain effects on the bacterial community structure of the fermentation pile. Mantel test further showed that there was a significant correlation between bacterial community structure and C/N (r=0.321, p=0.001), TN (r=0.265, p=0.003) and OM (r=0.278, p=0.002). These results showed that C/N played a key role in the nutrient content and bacterial community distribution of Myrica Rubra branch compost.
