香蕉钙调神经磷酸酶B亚基样蛋白基因MaCBL1的克隆及序列分析

摘    要:目的]为香蕉钙调神经磷酸酶B亚基样蛋白基因MaCBL1调控机理的研究奠定基础。方法]根据在香蕉果实抑制缩减文库中获得的一个CBL基因片段,采用PCR方法克隆了MaCBL1基因全长cDNA序列并对其进行了初步的生物信息学分析。结果]MaCBL1基因cDNA序列全长1 078 bp,编码213个氨基酸残基。多重序列比对结果显示,MaCBL1推导的氨基酸序列与其他植物来源的CBL基因的氨基酸序列同源性较高。遗传进化分析表明MaCBL1与其他植物如杨树、甘蓝、沙冬青、棉花和水稻的CBL基因的亲缘关系较近,并在同一个进化枝上。结论]该研究为进一步研究香蕉钙调神经磷酸酶B亚基样蛋白对香蕉生长发育、果实的成熟调控及对各种逆境反应的调控提供了依据。

Abstract:Objective] The aim was to lay the foundation for the study on the regulation mechanism of Calcineurin B-like protein gene from Musa acuminate(MaCBL1).Method] The cDNA sequence with full length of MaCBL1 gene was cloned by PCR according to the fragment of CBL gene obtained from suppression subtractive hybridization(SSH) library of M.acuminate fruit and its biological information was analyzed preliminarily.Result] The full length of the cDNA sequence from MaCBL1 gene was 1 078 bp,encoding 213 amino acid residues.The result of multiple sequence alignment showed that the deduced amino acid sequence of MaCBL1 shared higher homology with that of CBL gene from other plants.The phylogenetic analysis showed that the genetic relationship between the MaCBL1 gene and the CBL gene from other plants such as populus,Brassica oleracea,Ammopiptanthus mongolicus,Gossypium hirsutum and Oryza sativa was closer and they were in the same clade.Conclusion] The research provided the basis for the further study on the regulation of Calcineurin B-like protein on the growth and fruit maturation of M.acuminate and on the various stress responses.
