大叶黄杨内生真菌的分离及抑菌活性研究

摘    要:目的]从大叶黄杨中找到具有抗菌活性的内生真菌。方法]采用PDA双抗培养基从大叶黄杨茎和叶中分离内生真菌,对其分别进行液体培养,将发酵后所得的发酵液用乙酸乙酯萃取处理,以大肠杆菌、枯草芽孢杆菌、金黄色葡萄球菌为试验菌株,用纸片法对分离的真菌及其次生代谢产物进行了抑菌试验。结果]从大叶黄杨茎和叶中分离得到28株内生真菌,其中有21株内生真菌的发酵液粗提物对指示菌株有一定的抑制作用,占总分离菌株的75%。结论]大叶黄杨内生真菌的次生代谢产物中存在着丰富的天然活性抑菌物质,可为寻找新型抗菌物质提供资源。

Research on the Isolation of Endophytic Fungi from Euonymus japonicus and its Antibacterial Activity

Abstract:Objective] The endophytic fungi with antibacterial activity was found from Euonymus japonicus.Method] The endophytic fungi with antibacterial activity were isolated from Euonymus japonicus stem and leaf with dual-anti type of PDA medium and then cultured in liquid medium,respectively.The fermented broth was treated with ethyl acetatement.The E.coli,Bacillus subtilis,Staphylococcus aureus being used as the testing strains,the antibacterial test of the isolated fungi and their secondary metabolites to the testing strains were conducted with the disk diffusion method.Result] In 28 endophytic fungus strains isolated from the stem and leaf of Euonymus japonicus,the crude extract from the fermentation broth of 21 strains had the certain inhibition to the testing stain,accounting for 75% of the total isolated strains.Conclusion] There was the abundant natural antimicrobial active substance in the econdary metabolitesa of Euonymus endophyte,which would provide the new approach to find the resources with new type of antibacterial substance.
