肉仔鸡胸肌卫星细胞氧化应激模型的优化

摘    要:将体外培养的肉仔鸡胸肌卫星细胞(SC)按地塞米松(DEX)处理浓度(0、0.03、0.06、0.12、0.24、0.48和0.96 mg/mL)分为7组,分别测定SC的存活率(MTT法)、丙二醛(MDA)、氧自由基(ROS)、超氧化物歧化酶(SOD)和谷胱甘肽硫转酶(GST)含量或活性。筛选DEX处理的最佳时间和浓度以便优化肉仔鸡胸肌SC的氧化应激模型。结果表明,随DEX浓度升高,MDA和ROS产生量升高(P〈0.01),SOD与GST活性显著降低(P〈0.01)。随着DEX处理时间的延长,SC的存活率及GST和SOD活性均有所下降,MDA和ROS产生量上升。使用0.12 mg/mL的DEX处理SC 24 h后,SC存活率显著降低(P〈0.01)。说明DEX浓度为12 mg/mL处理24 h能使肉仔鸡骨骼肌SC产生适度的氧化应激,可以作为肉仔鸡胸肌卫星细胞的氧化应激模型。
