杜仲内生细菌对植物病原菌的拮抗作用

摘    要:为进一步丰富玉米病害的生防资源,通过平板稀释法,对健康杜仲组织进行内生细菌的分离纯化,共得到内生细菌55株, 55株内生细菌分别属于芽孢杆菌属(Bacillus)、类芽孢杆菌属(Paenibacillus)、伯克霍尔德菌(Burkholderia)、贪铜菌属(Cupriavidus)和赖氨酸芽孢杆菌属(Lysinibacillus)等5个属。利用平板对峙方法,筛选出对玉米小斑病和玉米茎基腐均有较好的抑制作用的内生细菌DZSY21,其抑菌率分别达到61.30%和79.10%,显微观察显示,拮抗菌株 DZSY21可引起病原菌菌丝断裂、畸形等现象。随后,对内生细菌DZSY21开展了gyrB及趋化性研究,结果表明,DZSY21为枯草芽孢杆菌,且对玉米根系分泌物具有较好的趋化性。

Abstract:To investigate the biological control agents to counteract the maize disease, plate dilution method was used to isolate the strains from the healthy Eucommia ulmoides. The results showed that the total of 55 strains of endophytic bacteria in Eucommia ulmoides were obtained, belonging to 5 genera: Bacillus, Paenibacillus, Burkholderia, Cupriavidus and Lysinibacillus. Then the antimicrobial effects of the endophytic bacteria were tested by dual culture, and the candidate DZSY21 was found to have significantly antagonistic effects on the growth and development of Bipolaris maydis and Fusarium moniliforme, resulting in the inhibitory rates of 61.30% and 79.10%, respectively. Microscopic examination of affected mycelia of phytopathogen showed some mycelia treated with the DZSY21 were swelled, contorted and broken. Molecular characterization assays indicated that the DZSY21 is Bacillus subtilis, containing chemotaxis to the root exudates of maize.
