引物原位标记技术及其应用

摘    要:本文综述了近几个来引物原位标记技术的发展和应用,并以检测猪染色体端位序列为例,介绍了这项技术的基本原理和实验程序。现已表明,引物原位标记技术不仅可以应用于检测染色体特定的DNA序列,而且可以应用于在细胞和组织切片上检测特定的RNA序列。

Abstract:Primed in situ labelling (PRINS) method has been developed as a fast and sensitive alternative to the more traditional in situ hybridization techniques. The strategy in PRINS technique is based on hybridization of synthetic unlabelled oligonucleotide(s) or short DNA fragment(s) to the complementary nucleic acid sequences in situ followed by primer extension in the presence of digoxigenin labelled or biotinlated nucleotides by a suitable polymerase. The labelled site of hybridization can be finally detected by specific antibodies such as anti digoxigenin Fab fragments or streptavidin. This overview summarized the development and applications of PRINS in the recent years. An example of PRINS detection of telomeric sequences (CCCTAA)n in porcine chromosomes was discussed, which presented an adapted protocol of PRINS experiment. Previous studies indicated that PRINS has many important applications not only in detection of specific sequences in inter /metaphase and polytene chromosomes, but also in visualization of RNA in situ in cellular and tissue preparations.
