广西地区甘蔗黄叶病病毒分子鉴定

摘    要:目的]为广西甘蔗黄叶综合征(YLS)的分子检测和抗病育种提供参考。方法]采集显症甘蔗叶样本,通过提取总RNA,利用引物P1/P2对样本递行反转录RCR(RT-PCR)扩增,对广西地区甘蔗黄叶病的病原体进行分子鉴定。结果]经引物P1/P2扩增出630 bp的目的片段。氨基酸和核苷酸序列分析结果表明,该片段是甘蔗黄叶病毒(SCYLV)外壳蛋白基因的一部分,与其他国家SCYLV分离物同源性达95%-99%。结论]广西甘蔗黄叶病病原确是SCYLV,该病毒可能是近年从区外或境外传入的。

Abstract:Objective] The reference for the molecular detection of sugarcane yellow leaf(YSL) and the disease-resisting breeding of sugarcane in Guangxi was provided through the research. Method] Total RNA, extracted from the sample with the disease symptom, was amplified with specific primers P1/P2 for RT-PCR. Result] A distinct fragment of about 630 bp was amplified.
