摘    要:从早熟甘蓝型油菜品种601的子叶中游离出原生质体,采用液体浅层培养,获得持续分裂的细胞。培养基中的2,4-D浓度对刺激细胞分裂至关重要。形成的小愈伤组织必须经继代培养(MS培养基补加2,4-D 0.2mg/L,KT 2mg/L)后,再转至MS分化培养基(补加NAA0.01mg/L,KT 3mg/L)上,才能分化出芽。分化的芽在MS生根培养基(补加IBA0.5mg/L)上,可以形成完整植株。

Abstract:Protoplasts of Brassica napus were isolated from cotyledon explant. Cell division took place and calli were obtained on thin-layer liquid medium. Sustained cell division was attributed to the presence of 2. 4-D in the medium. Young shoot regeneration occurred from the cotyledon-derived calli,which had been proliferated on the MS medium supplied with hormones of 2. 4-D 0. 2 mg/L and KT 2 mg/L.then being transferred onto a modified MS medium supplied with NAA 0. 01 mg/L.KT 3 mg/L. After the shoots were transferred onto the rooting medium supplied with IBA 0, 5 mg/L .intact plants were eventually produced.
