沙柳不同产地间遗传多样性的SSR分析

摘    要:分析沙柳种质资源的遗传多样性和遗传分化,为资源的有效利用、新基因的挖掘和新品种选育提供参考。应用SSR分子标记分析8个产地330份沙柳样本的遗传多样性。从45对SSR引物中筛选出19对多态性引物,共扩增出74条带,其中63条具有多态性,每对引物检测到等位位点数为2-11,平均多态性位点为3.89,平均有效等位基因数(NE)为1.8171,平均期望杂合度(He)为0.4079,平均观测杂合度(Ho)为0.3798,平均Shannon信息指数(I)为0.6447。利用POPGENE软件对SSR扩增结果分析表明,8个产地间的遗传相似度在0.9437-0.9908。UPGMA聚类分析表明,8个产地划分成3类;遗传分化程度和产地间的距离呈正相关。

Genetic Diversity of Salix psammophila from Different Producing Areas Analyzed by SSR Markers

Abstract:The purpose of this research was to investigate the genetic diversity and differentiation of cultivated Salix psammophila germplasm resources, and provide useful information on effective utilization of resources, new genes mining and breeding. In this study, 330 Salix psammophila in 8 areas were adopted to conduct genetic analysis by using SSR markers. Among 45 pairs of SSR primers, 19 pairs of primers were polymorphic. A total of 74 bands were amplified, in which 63 were polymorphic. The number of the allelic loci for each pair of primers was ranged from 2 to 11, and was 3.89 on average. The average effective number of alleles (NE), expected heterozygosity (He), observed heterozygosity (Ho), Shannon' s information index (I) were 1.8171, 0.4079, 0.3798, 0.6447 respectively. Based on the amplified SSR bands, genetic similarities among eight groups of Salix were varied from 0.9437 to 0.9908. were divided into 3 classes. The results showed Using POPGENE, cluster analysis showed that the 8 groups that there was a positive correlation between the genetic diversity and geographical origin.
