匈牙利速生型刺槐遗传转化再生体系的建立

摘    要:以匈牙利速生型刺槐无菌苗复叶叶柄为外植体,MS培养基为基本培养基,通过研究不同激素浓度组合对不定芽分化和生根的影响,建立了较好的遗传转化体系。结果表明,匈牙利速生型刺槐最适分化培养基为MS 6-BA1.0mg/L NAA0.5mg/L,不定芽再生率达77.5%;最佳生根培养基为1/2MS NAA0.2mg/L IBA0.1mg/L,生根率达100℅;10mg/L潮酶素可以抑制刺槐不定芽的诱导分化,5mg/L潮酶素可以抑制刺槐不定芽的生根。

Establishment of a Genetic Transformation System in Fast Growing Robinia pseudocacia from Hungary

Abstract:Adventitious shoots were induced from the raches of compound leaves of fast growing variety of Robinia pseudocacia introduced from Hungary in this study. The optimized medium for raches explants contains MS base medium combined with 6-BA 1.0 mg/L and NAA 0.5 mg/L, shoot regeneration rate is 77.5%. The medium for rooting of shoots is optimized as I/2MS base medium supplemented with NAA 0.2mg/L and IBA 0.1mg/L, with a rooting efficiency of 100%. Hygromycin at 10mg/L in the medium gives effective inhibition to the regeneration of shoots from the explants; in addition, hygromycin at 5mg/L in the medium shows obvious inhibition to the regeneration of roots from the shoots.
