甲醛法在玉米粗蛋白测定中的应用

摘    要:为改进凯氏定氮法的消化方式,探讨甲醛法测定玉米粗蛋白的可行性,分别用常规凯氏定氮浓硫酸消化法和过氧化氢浓硫酸消化法,以锥形瓶取代凯氏瓶,对玉米、玉米蛋白粉及玉米渣皮进行消化,消化后的样品分别以常规凯氏定氮法和甲醛法测定粗蛋白含量.结果表明,过氧化氢-浓硫酸混合液消化法,消化时间为15min,比常规方法(1h)缩短了45min,两种方法测定的粗蛋白变异系数分别为0.248%和0.378%,经t检验两种方法测定结果无显著差异,因此,甲醛法是一种简便、省时可靠的粗蛋白测定方法.

Abstract:The experiment was carried out to improve the digesting way of Kjeldahl method for nitrogen determination and research the application of formol-titration method on the determination of crude protein(CP) in maize.Maize,corn protein powder and corn lees were digested by Kjeldahl digestion and also digested by using hydrogen peroxide and concentrated sulfuric acid mixed solution as digesting reagents,with triangle bottle replacing for Kelvin nitrogen bottle.The assimilated samples were determined with Kjeldahl method and formol-titration method.The results indicated that the digesting time was reduced from 1 h to 15 min.The formol-titration method and Kjeldahl method coefficient of variation were 0.248 % and 0.378 % respectively.Two methods had almost the same results and not remarkable difference by t test(P>0.05).
