摘    要:为了得到一株降解PVA的芽孢杆菌,并对其进行固定化研究。利用PVA碘-硼酸平板法初筛,降解率测定法进行复筛,并通过正交试验探索其固定化工艺。经初筛得到8株可降解PVA的细菌,复筛出1株具有较高降解率的菌株3-9,48 h降解率为86.56%。该菌株鉴定为枯草芽孢杆菌(Bacillus subtilis subsp.subtilis)。该菌株的最佳固定化条件为:4%海藻酸钠,3%CaC12,包埋0.1 g的湿菌体,钙化时间4 h。结果表明固定化后的PVA降解菌比游离菌的降解性能提高了14.02%。本研究得到了一株高效降解聚乙烯醇的枯草芽孢杆菌,并确定其最佳的固定化条件,为PVA污水处理提供了菌种资源和理论基础。

Abstract:Screening of a strain with high tenor of PVA degradation Bacillus.Sixty bacterial strains were isolated from the soil samples and there were eight bacterial strains showed degrade activity to polyvinyl alcohol. A strainwith the highest degradation activity, 3-9 was obtained during the secondary screening.The degradation rate of 86.56.16S rDNA sequences analysis of strain 3-9 revealed that it had 100% similarity to that of Bacillus subtilis subsp. subtilis NR_027552. After the morphological, physiological and biochemical characteristics of this strain were further studied and compared with that of the type strain Bacillus subtilis subsp.subtilis NR_027552, strain 3-9 was finally identified as Bacillus subtilis subsp.subtilis.Optimal preparation conditions of the immobilized strains were carried out by the cross-test experiments. The best condition to 3-9 is to use sodium alginate of 4%, CaC12 of 3%, cells of 0.1g and to ealeify for4 hours. The immobilized cells and the freed cells degradation ability to PVA were studied. The results showed that the degrading ability of immobilized PVA degrading strains have greatly improved by 14.02% than free strains.
