构树叶粗多糖对环磷酰胺诱导免疫抑制小鼠免疫功能的影响

摘    要:旨在研究构树叶粗多糖预处理对环磷酰胺诱导免疫抑制小鼠免疫功能的影响。采用水提醇沉法提取构树叶粗多糖,用提取的构树叶粗多糖对小鼠进行15 d预防性给药,在给药的最后3 d,使用环磷酰胺诱导免疫抑制模型,采集血液检测各类白细胞数量、血清蛋白含量及血清抗体水平,采集脾脏组织和空肠组织观察病理变化。结果显示,与空白组相比,模型组小鼠的脾脏指数、白细胞、总蛋白、白蛋白、球蛋白、IgM、IgG均显著降低,模型组肠道黏膜组织受损,肠绒毛出现断裂脱落,脾脏组织的红髓区和白髓区界限模糊,白髓区的淋巴小结排列松散;与模型组相比,低剂量构树叶粗多糖(50 mg/kg)可显著提高小鼠血清中总蛋白、白蛋白、球蛋白、IgM、IgG的水平,白细胞、淋巴细胞、中性粒细胞数量也有增加,但差异不显著。综上所述,构树叶粗多糖可通过提升脾脏指数、白细胞数量、血清蛋白和血清抗体水平,改善肠道组织及脾脏组织损伤,从而增强环磷酰胺诱导免疫抑制小鼠的免疫功能,以低剂量的效果更佳。

Abstract:To investigate the immunological regulation of crude polysaccharide from Broussonetiapapyrifera leaves on cyclophosphamide-immunosuppressed mice. Water-extraction and alcohol-precipitation method was used to extract the the crude polysaccharide of Broussonetiapapyrifera leaves to chronically administer to mice for 15 days, and then the mice were intraperitoneal injected the cyclophosphamide to induce immunosuppressive model for 3 days. Blood samples were collected to detect the variation of white blood cells, the content of serum protein and serum antibody. Pathological of spleen and jejunum were observed with hematoxylin and eosin stain. The results showed that the spleen index, number ofwhite blood cells, content of total protein, albumin, globulin, IgM and IgG in the model group were significantly lower than that of control group. The damaged intestinal mucosa, broken intestinal villi could be observed in model group.Compared with the model group, the crude polysaccharide of Broussonetiapapyrifera leaf at low dose (50 mg/kg) could significantly increase the serum levels of total protein, albumin, globulin, IgM and IgG. Besides, the WBC, Lymp and Gran were also elevated by CPBL,but the difference was not significant. In conclusion, CPBL can improve the intestinal and spleen tissue damage, and enhance the immune function of cyclophosphamide- immunosuppressed mice.
