沼泥及其富集物中微生物多样性和区系变化的分析

摘    要:对从北京周边地区户用沼气池中采集的沼泥进行了富集培养及基因组DNA的提取,以古菌和细菌的通用引物PCR扩增16S rDNA基因的V3可变区,结合应用变性梯度凝胶电泳(DGGE)技术分析沼气池中微生物种群基因的多样性,以及富集前后的微生物群落区系变化,并回收优势的DNA片段进行序列分析。结果表明,微生物群落在沼气池的不同层次上呈现出空间分布多样性的差异,经过富集培养后,微生物种群的多样性及特异性发生了很大的差别。序列比对的结果显示,沼气池中的优势古菌菌群为甲烷鬃菌属(Methanosaeta)与甲烷螺菌属(Methanospiril-lum),细菌为梭菌目(Clostridiales)和互养菌属(Syntrophus)。经过富集培养后,古菌以甲烷八叠球菌属(Methanosarcinamazei)为主,细菌除保持了原有的优势菌群外,γ-变形菌纲(Gamma proteobacterium)也占有绝对的优势。
