茅莓总黄酮提取工艺的优化

摘    要:从茅莓(Rubus parvifolius L.)中提取总黄酮,采用吸光光度法测定提取液中总黄酮的浓度.在单因素试验的基础上采用L9(34)正交试验考察乙醇体积分数、料液比、超声处理时间及提取温度对总黄酮提取率的影响.结果表明,茅莓茎中总黄酮最佳提取工艺条件为乙醇体积分数30%、料液比1∶50(m∶V,g/mL)、提取温度80℃、超声处理时间90 min,此工艺条件下茅莓茎中总黄酮提取率为4.7 mg/g,此时叶中总黄酮的提取率高达12.2 mg/g,远高于根和茎中的.

Abstract:Total flavonoids were extracted from Rubus parvifolius L.,the concentration of flavones in extractant was detected by spectrophotometry.The effects of solid to liquid ratio,extraction time,volume fraction of alcohol solvent and extraction temperature were studied through L9(34) orthogonal test on the basis of single factor tests.The results showed that the optimum extraction conditions were,volume fraction of ethanol solution,30%;solid-liquid ratio,1∶50(m∶V,g/mL);extraction temperature,80 ℃;extraction time,90 min.Under these conditions,the yield of total flavonoids in R.parvifolius stem reached 4.7 mg/g.The extraction yield of total flavonoids in R.parvifolius leaf was 12.2 mg/g,much higher than that in the stem and root.
