摘    要:采用单因素和正交试验相结合,对笔者分离保存的生防放线菌淡紫灰链霉菌(Streptomyces lavendu-lae)xjy菌株的液体发酵培养基和培养条件进行了优化。单因子试验结果表明,碳源葡萄糖、麸皮和空白处理三者之间无显著差异;氮源为豆粕、鱼粉和黄豆粉时发酵效价明显高于空白处理和其他氮源;添加0.04%的磷酸氢二钾和0.01%的硫酸亚铁可促进发酵液活性物质的产生。正交试验得出较适合于工业生产的摇瓶培养基配方为:麸皮2%、豆粕1%、氯化钠1.5%、碳酸钙0.6%、硫酸铵1%、磷酸氢二钾0.04%、硫酸亚铁0.001%。优化后的发酵液效价提高了36%。较为适宜的发酵非营养条件为:最适温度为20~35℃、摇床转速为150 r/min、初始pH值为5.5~8.0、装料量为50 mL/250 mL。这些结果为该菌株今后的应用、抗生素的分离提纯和产业化提供了依据。

Abstract:Active components in fermentation of the Streptomyces lavendulae strain xjy which separated from soil in our lab showed antagonism against lots of pathogen.The optimum fermentation conditions of strain xjy were optimized in the stud,The results of single fact or experiment showed no significant differenceing lucose,wheat bran and blank as carbon source and so ybean meal was the best nit rogen source.K2HPO4(0.04%) and FeSO4·7H2O(0.001%) were increased active components in fermentation.The orthogonal test showed that the optimum medium was wheat bran 2%,soybean meal 1%,NaCl 1.5%,CaCO3 0.6%,(NH4) 2SO4 1%,K2H PO4 0.04% and FeSO4·7H2O 0.001%.The active components in fermentation production was increased by 36% when the strain xjy was cultivated in the optimum medium instead of the initial one.The higher yield of active components was obtained on the conditions that the temperature is at 20~35,the shaker rotation speed is at 150 r/min,the medium capacity is 50 mL in 250 mL flask,the initial pH value is 5.5~8.0.
