皂荚皂苷元制备及纯化工艺研究

摘    要:为优化皂荚皂苷酸解制备皂苷元的工艺,以皂荚皂苷粗品为原料,考察酸浓度、酸解时间、酸解温度、料液比和酸种类对皂荚皂苷酸解转化成皂荚皂苷元的影响。结果表明,最佳酸解工艺为盐酸浓度4 mol/L、酸解时间4 h、酸解温度80℃、料液比1:30(m:v),采用优化后的酸解条件能够得到含量在65%左右的皂荚皂苷元产品。再通过有机溶剂萃取法和重结晶法对酸水解后的皂苷元进行纯化,确定最佳的萃取剂为三氯乙烯,料液比为1:20(m:v),萃取时间在90 min内变化不大,萃取1次,采用60%乙醇重结晶,得到的总皂苷元纯度可达76.31%。

Abstract:To optimize the preparation process of sapogenin from Gleditsia sinensis saponin by acid hydrolysis, the effects of acid concentration, acidolysis time, acidolysis temperature, material-to-liquid ratio and acid types on the conversion of saponin into sapogenin by acid hydrolysis were investigated. The results showed that the optimum conditions were as follows: hydrochloric acid concentration of 4 mol/L, hydrolysis time of 4 h, hydrolysis temperature of 80℃, solid-liquid ratio of 1:30 (m:v), and the content of Gleditsia sinensis sapogenin product was about 65%. The sapogenin after acid hydrolysis was purified by organic solvent extraction and recrystallization. The best extraction agent was trichloroethylene, the solid-liquid ratio was 1:20 (m:v), and extraction time did not change much within 90 minutes. After one extraction and recrystallization with 60% ethanol, the purity of total sapogenin could reach 76.31%.
