摘    要:目的]探讨影响钼锑抗比色法进行磷细菌发酵液中有效磷含量测定的因素。方法]以巨大芽胞杆菌为出发菌株,采用摇瓶发酵法培养有机磷细菌,用钼锑抗比色法测定上清液中有效磷含量,研究超声波处理,发酵体积、活性碳、反应液pH值和发酵液离心等因素对钼锑抗比色法测定有效磷含量的影响。结果]反应液最大吸收波长在720 nm左右;发酵体积、加入活性碳与否、反应液的pH值等对测定结果影响不大,在筛选时可大大提高效率;超声波处理20 min效果明显;在用50 ml容量瓶进行定容时,取样体积以2.5 ml为宜;对发酵液进行离心效果更好些,但在必要的情况下,可静置后取上清测定。结论]该研究为建立有机磷细菌诱变筛选体系提供理论依据。

Abstract:Objective] The research aimed to discuss the factors that affected the determination of available P content in the fermentation liquid of P bacteria by Mo-Sb colorimetry.Method] With Bucillus megaterium as the original strain,the organic P bacteria was cultured by shaking flask fermentation method and the content of available P in the supernatant was determined by Mo-Sb colorimetry.And the effects of such factors as ultrasonic treatment, fermentation volume,activated carbon,pH value of reaction solution and the centrifugation of fermentation liquid on the determination of available P content in the fermentation liquid of P bacteria by Mo-Sb colorimetry were studied.Result] The maximum absorption wavelength of reaction solution was about 720 nm. The fermentation volume,adding the activated carbon or not,pH value of reaction solution and so on had no great effect on the determination results and they could increase the screening efficiency.The effect of ultrasonic treatment for 20 min was obvious.When 50 ml volumetric flask was used,the suitable sampling volume was 2.5 ml.It was better to centrifugate the fermentation liquid and the supernatant could be taken after standing in the determination when necessary. Conclusion] This research provided the theoretical basis for establishing the mutation screening system of organic phosphorus bacteria.
