台湾过山香组培快繁技术研究

摘    要:以当年生台湾过山香带腋芽半木质化茎段作为外植体,开展组织培养试验研究,建立组培快繁体系。结果表明:最佳外植体诱导培养基为1/2MS+6-BA 1.0 mg/L+NAA 0.1 mg/L,诱导率可达89.7%;最佳增殖培养基为DCR+6-BA 0.8 mg/L+NAA 0.1 mg/L,增殖率达5.24倍,继代周期为35 d,不定芽生长状况好;最佳生根培养基为1/2MS+IBA 0.5 mg/L+ABT 1生根粉0.4 mg/L,生根率达95.26%;以泥炭土:黄心土:珍珠岩:钙镁磷肥(3.0:2.0:1.0:0.2),移栽成活率达93.7%,且7 d缓苗,茎秆木质化迅速,茎《浓绿,长势好。

Study on Tissue Culture Techniques for Clausena excavata Burro. f.

Abstract:Stems with axillary buds from Clausena excavata Burm. f. were used as explants for tissue culture and tissue culture was establishment. The results showed that the suitable medium of explants inducing was 1/2MS supplemented with 6-BA 1.0 mg/L and NAA 0.1 mg/L,and the inducing rate was 89.7%.The DCR supplemented with 6-BA 0.8 mg/L and NAA 0.1 mg/L was the optimized proliferating medium with multiplication coefficient being 5.24 and the growth cycle being 35 d ,and the shoots grew well. The best rooting medium was 1/2MS supplemented with IBA 0.5 mg/L and ABT 1 rooting power 0.4 mg/L,and the rooting rate was 95.26%.The plantlets were transplanted in mixture media with peat ,yellow soil and perlite calcium magnesium phosphate (3.0:2.0:1.0:0.2), and the survival rate was 93.7%.And 7 d later, the seedling was survival, stem was quickly lignification, leaf was deeper green and growing well.
