摘    要:目的]研究不同染色剂对SDS-PAGE染色体系的影响.方法]通过SDS-PAGE染色体系中不同考马斯亮蓝染料、染色方法和染色时间的比较,探讨适合油棕和椰子的最佳染色体系.结果]改进后的方法染色效果好,较好脱色,凝胶背景清晰,容易分析蛋白含量较低的条带.染色时间短(2 h),条带较少,一些低丰度的蛋白条带未能清晰地显示出来,适当延长染色时间(6h或过夜)效果较好.3种不同染料均能较好地对蛋白质条带染色,R350和R250相比G250效果稍好些,条带着色深,蛋白质含量低的细小条带也能较好的染色.结论]SDS-PAGE是开展蛋白质组学研究的关键技术,建立简易、稳定的染色方法具有重要意义.

Abstract: Objective] To study effects of different dyes on SDS-PAGE gel staining system. Method] Different SDS-PAGE staining methods with different Coomassie Brilliant Blue dye and staining time were compared to explore suitable staining system for oil palm and coconut SDSPAGE study. Result] The improved method of dyeing effect is good, good decolorization, gel background is clear, easy to analysis with low protein content. Short dyeing time (2 h), with less band, some low abundance protein band failed to show, prolonged dyeing time (6 h or overnight) would be better. Conclusion] SDS-PAGE is a key technology to carry out proteomic studies, the establishment of a simple, stable staining method is important.
