摘    要:目的]为了建立优化的根癌农杆菌介导的烟草外源基因转化体系,提高烟草外源基因转化率。方法]以无菌烟草苗叶盘预培养时间和农杆菌与烟草叶盘的共培养条件为试验要素,筛选适宜的预培养时间和共培养条件范围,其余操作按常规步骤进行。结果]烟草叶盘的预培养适宜时间为27℃下35~40h;农杆菌与烟草叶盘共培养的适宜条件为22~24℃下共培养30~45h,其中23℃下共培养45h效果最佳;之后转入选择培养基内培养。结论]该研究可为烟草外源基因的转化提供理论依据。

Abstract:Objective] In order to establish optimized Agrobacterium tumefacien-mediated transformation system for exogenous genes of tobacco.Method] The pre-culture time of tobacco explants and the co-culture conditions of Agrobacterium tumefacien-tobacco leaf disc was chose as testing elements,the remaining operating made by usual steps.Result] Experimental results showed that the appropriate pre-culture time of tobacco leaf disc was 35-40 h under 27 ℃.The optimal co-cultured conditions of Agrobacterium tumefacien-tobacco leaf disc was 30-45 h under 22-24 ℃,and the best condition was 45 h under 23 ℃,then cultured within selection medium.Conclusion] The study can provide theoretical basis for transformation of exogenous gene of tobacco.
