普通小麦新品种陕农33未成熟胚高频再生体系的建立

摘    要:陕农33是2012年审定的一个高产、稳产、强筋优质的小麦优良新品种。为了解陕农33组织培养特性,以科农199、H261为对照,研究陕农33未成熟胚愈伤组织的诱导和分化能力以及AgNO3、CuSO4对其组织培养的影响。结果表明:陕农33未成熟胚愈伤组织诱导率为98.11%、褐化率为2.48%、再生分化率为80.23%。差异显著性分析显示:陕农33与对照科农199(诱导率99.43%、褐化率2.25%、分化率71.18%)没有显著性差异(P0.05),但显著(P0.05)高于H261(诱导率96.24%、褐化率7.81%、分化率62.24%);说明陕农33具有较好的组织再生能力。另外,在培养基中添加4.0mg/L的AgNO3可以进一步提高愈伤组织诱导率至99.41%、分化率至82.35%,使褐化率降至1.68%;添加2.50mg/L的CuSO4可以提高绿苗分化率至84.17%。

Abstract:Shaannong 33 authorized in 2012 is a new variety of wheat with high and stable yield, strong gluten quality. To understand the tissue culture characteristics of Shaannong 33 , Kenong199 and H261 were served as control to analyze induction and differentiation ability of immature embryo callus of Shannong33 and the impact of AgNO3, CuSO4 on the tissue culture. The results showed that: callus induction rate of Shaannong 33 immature embryos was 98.11%, callus browning rate was 2.48%, and differentiation rate was 80.23%. Shaannong 33 had no significant difference (P> 0.05) with Kenong199 (induction rate of 99.43%, browning rate of 2.25%, differentiation rate of 71.18%), but significantly different (P<0.05) to H261 (induction rate of 96.24%, browning rate of 7.81%, differentiation rate of 62.24%); which suggested that Shaannong 33 has better tissue regeneration ability. In addition, 4.0 mg/L of AgNO3 added in the culture medium can further improve the callus induction rate to 99.41%, the differentiation rate to 82.35%, reduce the browning rate to 1.68%, and adding 2.50 mg/L CuSO4 can improve the green plant regeneration rate to 84.17%.
