紫芝子实体多糖的特征分析及其体外免疫活性

摘    要:通过乙醇分步沉淀法从紫芝(Ganoderma sinensis)子实体水提取物中分离多糖,得到分步醇沉的粗多糖样品30E、50E和70E,比较了3个粗多糖样品的得率、多糖含量、单糖组成、重均分子量分布特征、刺激RAW 264.7细胞释放NO和促进小鼠脾淋巴细胞增殖的活性.结果表明:30E、50E和70E的多糖得率均较低,分别为0.04%、0.17%和0.49%;水解后的单糖种类基本相同,均有岩藻糖、半乳糖、葡萄糖和甘露糖,但是单糖比例差异较大,70E中还含有3.4%的木糖;紫芝多糖的重均分子量分布范围较广,从1.450×104到1.276×107,30E中有2个大分子量组分(重均分子量分别为8.867×106和1.276×107),50E和70E的重均分子量较小(<2×104);分步醇沉的各粗多糖样品均具有体外刺激巨噬细胞释放NO和促进小鼠脾淋巴细胞增殖的活性,其中30E的活性最好.

Properties and in Vitro Immunological Activities of Polysaccharides from Ganoderma sinense Fruit Bodies
Abstract:One kilogram of Ganoderma sinense fruit bodies were suspended in double-distilled (dd) water (solid∶liquid ratio,1∶20 g∶mL) and extracted at 100 ℃ for two hours.After filtration,the solid residue was extracted again for one hour and the combined filtrates were concentrated under vacuum to 1 L and centrifuged to remove any precipitation.Absolute ethanol was added to a final concentration of 30%,and the solution was stored at 4 ℃ for 12 h.Precipitated material was collected by centrifugation,washed once with 30% ethanol and re-dissolved in dd water.After removing residual ethanol using a 100 ℃ water bath,the sample was freeze-dried to yield the crude polysaccharide fraction 30E.Two further fractionation steps using 50% and 70% ethanol concentrations were carried out on the supernatant from the first fractionation to yield crude polysaccharide fractions 50E and 70E respectively,and the yields,polysaccharide contents,monosaccharide compositions and in vitro immunological activities of all three fractions were evaluated.Yields of 30E,50E and 70E were 0.04%,0.17% and 0.49%,respectively.Monosaccharide compositional analysis revealed that all three crude polysaccharides contained fucose,galactose,glucose and mannose,and small amounts of xylose (3.4%) were also detected in fraction 70E.HPSEC-MALLS-RI (high-performance size exclusion chromatography multi-angle laser light scattering-refractive index detector) analysis indicated that the molecular weights of the G.sinense polysaccharides ranged from 1.450 × 104 to 1.276 × 107.Two high molecular weight fractions (Mw 8.867 × 106 and 1.276 × 107,respectively) were present in 30E,whereas 50E and 70E contained fractions of relatively low molecular weight (<2 × 104).All three crude polysaccharide preparations,especially 30E,stimulated the release of NO by macrophages and the proliferation of mouse lymphocytes.
