茶树基因组DNA提纯与鉴定

摘    要:采用在细胞核被裂解之前去除细胞质中的茶多酚和蛋白质,而后用SDS裂解细胞核,异丙醇和乙醇沉淀基因组DNA的方法,分别从不同茶树品种新梢、干梢及冰冻新梢中成功地提取和纯化基因组DNA,并对其DNA的得率和质量进行了鉴定。DNA的得率在205~963ng/mg鲜重之间,所得到的DNA样品片段均大于21kb,适宜于进行限制性酶切和RAPD反应。实践证明,上述提取富含酚类物质植物基因组DNA的方法,不仅迅速简单,而且经济有效。

Abstract:Polyphenols and protein in the cytoplasm were removed before the nucleus was decomposed by SDS, and the genomic DNA was isolated and purified from fresh, dry and frozen shoots by precipitating the genomic DNA with isopropanol and ethanol. Based on the determination of yielding and fragment length, the yield of the DNA by the method was 205—963 ng/mg·FW, and the length of the DNA fragment was more than 21 kb. The isolated DNA could be directly used for RFLP and RAPD analyses. The isolating method was proved to be a fast, easy and economic one for isolating genomic DNA from plants containing high polyphenols.
