摘    要:大豆小复叶外植体在含NaCl25、50、75和100mmol/L并加Ca(NO3)210mmol/L的培养基中,大豆离体胚在含NaCl50和100mmol/L并分别加Ca(NO3)2、10、15和20mmol/L的培养基中进行耐盐筛选培养。实验结果表明,在盐胁迫下加Ca有缓解盐害的效应.小复叶外植体在较高浓度盐胁迫下的出愈率和根、芽分化率增高;对离体胚在盐胁迫下的生长有促进作用,NaCl50盐胁迫下以Ca15效果最佳。NaCl100盐胁迫下,以Ca20效果较好.在盐胁迫下加Ca可减少离体胚再生植株中Na+和Cl-的积累,而增加Ca+和K+的积累,并使MDA含量减少.因而有减轻Na+和Cl-毒害和膜脂过氧化的作用。

Abstract:Soybean young compound leaves as explants were cultured on themedia containing 20, 50, 75 and 100 mmol/L NaCl with vitrol were cultured on themedia containing 50 and 100 mmol/L NaCl with 10,15 and 20 mmol/L Ca (NO3)2respectively for screening salt-tolerant plantlets. The results showed that Ca had an effect on relieving salt injury of soybean under salt stress. Under the higher salt stress, Ca increased the callus forming rate and differentiation rate of root and bud of the compound leaf explants. It also promoted the growth of embryo in vitro under salt stress, with Ca 15 is the best diminished the accumulation of Na and Cl- increased that of Ca and K , diminished MDA content of plantlets cultured from embryo in vitro. It indicated that Ca has an effect on relieving injury of Na and Cl-, and relieving membrane lipids superoxidation.
