西藏产弹性蛋白酶的枯草芽孢杆菌分离及鉴定

摘    要:目的]为了从西藏林芝分离出产弹性蛋白酶的枯草芽孢杆菌。方法]根据西藏林芝的实际,从西藏林芝八一镇的屠宰场附近的土壤、污水中采样和30℃培养分离。以水解弹性蛋白形成透明圈直径与菌径之比最大的菌株作为研究对象,对弹性蛋白和酪蛋白平板水解的透明圈直径和菌径进行测量,对其个体形态、菌落特征、生理生化特征方面进行鉴定。结果]据个体形态、菌落特征、生理生化特征,所分离出的菌株鉴定为枯草芽孢杆菌。在弹性蛋白和酪蛋白平板上,菌株的平均HC分别为1.64和2.57。该菌产弹性蛋白酶、淀粉酶、过氧化氢本酶,分解酪蛋白的酶。结论]该研究为保藏西藏特有的产弹性蛋白酶的枯草芽孢杆菌奠定基础。

Abstract:Objective] The research aimed to isolate Bacillus subtilis producing elastase in Nyingchi of Tibet.Method]According to the condition of Nyingchi Tibet,taking the example in the vicinity of the slaughterhouse's soil and sewage of Bayi Town in Nyingchi,it was isolated by incubating at the temperature of 30 ℃.The strain whese ratio(HC) of hydrolysis of elastin formed transparent circle diameter to the diameter of the colony was the largest was taken as research object.Transparent circle diameter and the diameter of the clony were measured in the plates hydrolysis of elastin and casein.It was identified in term of individual morphology,colony characteristics,physiological and biochemical characteristics.Result] The strain of bacteria which was isolated was identified as Bacillus subtilis according to the morphologic,physiological and biochemistric characteristics.Average HC values were respectively 1.64 and 2.57 on the plates hydrolysis of elastin and caseine.The strain could produce elastase,amylase,catalase and the enzyme which could hydrolyse casein.Conclusion]The research could provide the basis for the preservation of Tibet special Bacillus subtilis producing elastase.
