青海栽培黄管秦艽的叶绿体DNA psbA-trnH核苷酸变异和遗传分析

摘    要:目的]研究青海栽培黄管秦艽(Gentiana officinalis H.Smith)的遗传多样性,对黄管秦艽遗传分化进行分析,为其品种选育提供建议。方法]应用叶绿体DNApsbA-trnH序列对青海地区栽培黄管秦艽6个居群进行遗传多样性分析。结果]共发现10种不同的单倍型,通过单倍型序列间比对,发现了7个变异位点,黄管秦艽具有较高的遗传多样性(h=0.771),单倍型多态性水平(h)为0.563~0.857,核苷酸多态性水平(π)为0.002 43~0.006 31。居群遗传分化系数Gst为0.196 0,基因流Nm值为2.05,80.40%遗传变异主要存在于居群内。结论]青海栽培黄管秦艽的种质遗传多样性较高,有利于培育高品质的药材。

Study on the Chloroplast psbA-trnH Nucleotide Variation and Genetic Differentiation in Cuitivated Plants of Gentiana officinalis from

Abstract:Objective] The aim was to investigate the genetic diversity of main germplasm resources of Gentiana officinalis H.Smith in Qinghai and the genetic differentiation of the germplasm.Method] The cpDNA of psbA-trnH gene was sequenced to analyze the genetic diversity of six populations of G.officinalis.Result] A total of 10 distinct haplotypes were detected in the studied populations,and 7 variable sites were found by comparing their sequences.G.officinalis(h=0.771) possessed high level genetic diversity.Genetic diversity largely varied within populations,ranging from 0.563 to 0.857 for haplotype diversity,and from 0.002 43 to 0.005 83 for nucleotide diversity,respectively.Genetic differentiation among populations(Gst) of G.officinalis was 0.196 0;gene flow(Nm) was 2.05;80.40% of the genetic variability existed in population.Conclusion] The cultivated G.officinalis in Qinghai showed rich genetic diversity,which is good for the production of high quality herb medicine.
