苯酚-硫酸法测定灰树花中多糖含量的研究

摘    要:目的]建立一种快速、稳定的适于测定灰树花多糖含量的方法。方法]热水浸提法提取灰树花多糖,采用苯酚-硫酸法,以葡萄糖为标准品,以蒸馏水为空白对照,于490 nm波长处分别测定对照品和供试品溶液的吸光度,按标准曲线计算葡萄糖含量。结果]葡萄糖在20~170μg/ml范围内线性关系良好,回归方程为Y=0.009 7X-0.096 4(Y为吸光度,X为葡萄糖浓度,μg/ml),相关系数r=0.998 5,低、中、高3个不同浓度的加样回收率分别为101.19%、100.01%和100.23%(n=5),其RSD值分别为1.20%、0.85%和0.55%。结论]苯酚硫酸法是一种快速、稳定、重现性好的灰树花多糖含量测定方法。

Determination of Polysaccharide Content in Grifola frondosa by Phenol-Sulferic Acid Method

Abstract:Objective] To develop a rapid and stable method for determining the polysaccharide content in Grifola frondosa.Method] Polysaccharide was extracted from Grifola frondosa by the boiling-water method.The content of polysaccharide in Grifola frondosa was determined by the phenol-sulferic acid method,with the glucose was as reference standard and the distilled water as the blank control,the absorbance of the control and test solution was determined at 490 nm and the content of glucose was calculated by using the standard curve.Result] Glucose showed good linear relation in the range of 20-170 μg/ml and the regression equation was Y =0.009 7 X-0.096 4(Y and X stood for absorbance and concentration of glucose respectively),the correlation coefficient was 0.998 5.The average recoveries for three different concentrations were 101.19%,100.01% and 100.23%(n=5),and the relative standard deviations were 1.20%,0.85% and 0.55%,respectively.Conclusion] The method was rapid and stable with good reproducibility.It could be used to determining the content of polysaccharide in Grifola frondosa.
