禾谷镰刀菌蛋白激酶FgSid1的功能研究

摘    要: 实验室前期已经对禾谷镰刀菌中116个蛋白激酶进行了鉴定,发现FgSID1基因(FGSG_07344)敲除突变体在有性生殖过程中存在严重缺陷,但其在禾谷镰刀菌中的具体功能却未被详细报道。本研究通过表型分析和亚细胞定位等实验明确了FgSid1在营养生长、有性发育和致病性等方面的功能。本研究发现FgSID1基因敲除突变体菌落生长速度显著变慢,分生孢子隔膜数量减少且产孢量下降;在有性生殖阶段,FgSID1基因敲除突变体的子囊壳形态和大小均正常,但子囊畸形且子囊孢子形成有缺陷;此外,FgSID1基因敲除突变体只能侵染接种小花,而无法扩展到穗轴及临近的小穗,且接种点的小麦籽粒DON毒素含量显著下降;研究还发现FgSID1基因敲除突变体的分生孢子、营养菌丝和子囊孢子的分隔中均含有多个细胞核;本研究又通过亚细胞定位实验发现FgSid1定位于微管组织中心。因此,蛋白激酶FgSid1可能通过参与禾谷镰刀菌无性和有性阶段的细胞分裂影响禾谷镰刀菌的营养生长、分生孢子和子囊孢子的发育以及侵染能力。

Abstract:According to previous studies in our laboratory, a total of 116 protein kinases were identified in Fusarium graminearum and FgSID1 (FGSG_07344) was described to play a role in the sexual reproduction. However, the function of FgSID1 was still not clear in F. graminearum. In this study, the function of FgSid1 was further clarified through phenotypic analysis and subcellular localization. The results showed that deletion of FgSID1 affected the vegetative growth rate, conidia production, asci and ascospore formation, virulence and deoxynivalenol (DON) production. Furthermore, the Fgsid1 mutant had multiple nuclei in the compartment of vegetative hyphae, conidia and ascospore. In addition, FgSid1-GFP was located at the microtubule organizing centers (MTOC). Taken together, protein kinase FgSid1 was important for fungal growth, virulence, conidial and sexual reproduction, which may due to its function in cell division during the asexual, sexual and infectious growth of F. graminearum.
