摘    要: 甜瓜坏死斑点病毒(melon necrotic spot virus,MNSV)是侵染甜瓜的重要病毒之一,为害严重。为了明确MNSV的致病机制,本研究以pGBK-p42为诱饵载体,利用Mating的方法从感染MNSV的甜瓜cDNA文库中筛选出6个与p42互作的寄主因子,进一步利用酵母双杂交系统、双分子荧光互补技术(Bimolecular fluorescence complementation,BiFC)等验证p42与寄主因子3-羟基异丁酸脱氢酶(3-Hydroxyisobutyrate dehydrogenase-like 1, 3-HIBADH)的互作,并明确了p42 蛋白的s-domain是与3-HIBADH互作的关键区域。此外,利用激光共聚焦显微镜观察发现p42 s-domain与3-HIBADH作用改变了3-HIBADH蛋白在烟草表皮的亚细胞定位,进一步证实p42可与3-HIBADH发生相互作用。该结果为阐明甜瓜响应MNSV侵染的互作机制奠定了基础。

Abstract:Melon necrotic spot virus(MNSV)is one of the main viruses infecting melon in China.Little is known about the mechanism of interaction between MNSV and the host factors. In this study, we screened the melon host factors interacting with p42 of MNSV and further verified the interaction between them. The bait vector pGBK-p42 was constructed to screen the host factors, the results showed that six host factors could interact with p42 , the interaction between p42 and host factor 3-Hydroxyisobutyrate dehydrogenase- like 1(3-HIBADH) was further confirmed by yeast two-hybrid system. The specific interaction between p42-s domain and 3-HIDBADH was confirmed by yeast two-hybrid system, BiFC and co-localization assays.The results of gene co-localization showed that the location of the 3-HIBADH gene was changed because of the interaction with p42-s domain. A number of small particles were formed in the cytoplasm which further indicated that p42 interacted with 3-HIBADH. The results could promote further understanding the mechanism of interaction between host factors and MNSV.
