摘    要:采用PCR产物直接测序技术对重庆地区117群东方蜜蜂样品的线粒体DNA细胞色素b基因(Cyt b)420 bp长度序列进行了分析。结果显示:在所得到的序列中共检测到11个核苷酸多态位点,其中单一变异位点2个,简约信息位点9个,序列突变率为2.62%,序列T,C,A,G平均含量分别为44%,14.2%,32.9%,8.9%,共定义8种单倍型,单倍型多样度为0.773±0.021,核苷酸多样度为0.003 82±0.000 32,单倍型网络关系图揭示重庆地区东方蜜蜂群体没有明显的遗传分化。

Genetic diversity of Apis cerana in Chongqing Using mtDNA Cytochrome b gene Sequences
Abstract:The sequences of mtDNA Cytochrome b gene of 120 colonies of Apis cerana collected from Chongqing area were analyzed by the method of PCR with direct se- quencing. The results showed: A total of 8 mitochondrial haplotypes with 1 1 polymorphic sites included 2 singleton polymorphic sites and 9 parsimony informative sites were detected in the samples. The average frequencies of T, C, A and G for the sequences of Apis cerana were 44%, 14.2%, 32.9% and 8.9%, respectively. And haplotype diversity, nu- cleotide diversity, between two different regions were 0.773 ±0.021 and 0.003 82±0.000 32. There was no obvi- ously genetic differentiation in Chongqing indicated by the Network relation.
