苦参药材中总生物碱含量的测定

摘    要:目的]用酸性染料比色法测定不同产地苦参药材中总生物碱的含量。方法]苦参总生物碱的浓度50%乙醇溶液,加一定pH值的缓冲溶液溴麝香草酚蓝指示液振摇后,氯仿萃取,萃取液于一定波长处测定吸光度A。结果]氧化苦参碱检测浓度在0.013~0.026mg/ml范围内与吸光度线性关系良好(r=0.997 1)。用氧化苦参碱作为对照品,在pH值7.0的条件下加入2.0 ml的溴麝香草酚蓝溶液,在417 nm处有最大吸光度,该条件下可准确地测定苦参药材中总生物碱的含量。试验测得不同产地苦参药材中总生物碱含量差异较大,其中所购得的陕西、甘肃苦参药材中总生物碱含量较高,分别为1.801、1.778 mg/kg。结论]该试验所采用的方法操作简便、准确,可通过总生物碱的含量客观全面地评价苦参药材的质量。

Comparison of Total Alkaloid Content in Sophora flavescens Ait.Medicinal Materials from Different Producing Areas
Abstract:Objective] The research aimed to determine the content of total alkaloid in Sophora flavescens Ait.medicinal materials from different producing areas by acid dye colorimetry.Method] Buffer solution with certain pH value and bromthymol blue indicator were added to 50% ethanol solution of total alkaloid from S.flavescens.After oscillation and chloroform extraction,the absorbency(A) of extract liquid was determined at certain wavelength.Result] The detection concentration of oxymatrine in the range of 0.013-0.026 mg/ml had a good linear relationship with its absorbency(r=0.997 1).With oxymatrine as control substance,2.0 ml bromthymol blue solution was added at pH value of 7.0.It had the maximum absorbency at eh wavelength of 417 nm.Under these conditions,the content of total alkaloid in S.flavescens medicinal materials could be determined accurately.It was known from the test determination results that the content of total alkaloid in S.flavescens had greater difference among different producing areas.The content of total alkaloid in S.flavescens medicinal materials bought from Shaanxi and Gansu were higher,being 1.801 and 1.778 mg/kg respectively.Conclusion] The used method in the test was simple and accurate.And the quality of S.flavescens medicinal materials could be evaluated objectively and roundly through the content of total alkaloid.
