摘    要:比较5个供体的心叶和幼茎愈伤组织在 MS 和 N_6培养基中诱导分化绿苗过程 RNA,DNA 和可溶性蛋白质的动态变化.结果发现,在绿苗诱导分化的第6—9 d,核酸总量和 RNA含量均达到最高值,第9 d 后两者逐渐下降.DNA 含量从诱导分化开始即呈下降趋势,第9d 时DNA 含量最少,然后义逐渐增加.蛋白质含量在诱导分化的前3 d 增加显著,第3 d 后呈下降趋势,第9 d 后渐趋减缓.不同的培养基对生物大分子的含量变化有一定影响.本试验揭示了在甘蔗愈伤组织诱导分化绿苗过程中,生物大分子变化的不同阶段与细胞分化和器官形成的关系.

Abstract:In this paper,the different sugarcane explants obtained form the young leaf, young internode of 5 varieties were used to investigate the differences of callus differentiation and plant regeneration in MS and N6 media.The results showed that the contents of nucleic acid,RNA,DNA and water soluble protein were different during 18 days of plantlet regener- ating from different calli.The nucleic acid total content of callus was found to increase during 18 days of plantlet regeneration,6-9 days after callus subculture the content appeared a peak, then declined gradually with green dot emergement.The change of RNA content was similar to the nucleic acid total content during the plantlet regeneration.The chage of DNA content of callus in two media was significant,but 9 days after subculture the DNA content was lower than other stages.The water soluble protein content of clutures increased gradually during the plantlet regeneration,3 days after subculture the content appeared a peak,then declined.The above biochemical analysis results suggested that the changes of these metabolites were relat- ed to the plantlets regeneration.
