摘    要:以山新杨(24年生)优树的2年生萌生枝上的叶片为材料,进行了不同培养基及不同影响因子对山新杨叶片不定芽再生能力影响试验,结果表明:山新杨叶片再生的最适合培养基是1/2MS+BA0.5 mg.L-1+NAA0.1~0.2 mg.L-1,再生频率达100%,叶片培养的最佳取材部位是树枝茎尖展叶开始第1~3片叶;叶片正面向上或向下接种的培养基,对其形成不定芽无显著影响。

Abstract:Taking leaves of 2-year-old stump plant of selected tree of Populus davidiana×P.bolleana(24-year-old) as materials ,tests of different culture medium and affectoi on regeneration capacity influence of adventitious bud of leaves of Populus davidiana×P.bolleana were conducted.Results showed that the optimal culture medium is 1/2MS+BA0.5 mg·L~(-1)+NAA0.1~0.2 mg·L~(-1).Regeneration frequency reaches 100%.The best parts taken from leaves are on leaf wxpansion(from the first leave to the third leave) of stem apex of branches.Culture medium inoculated upward and downward the pressure side of the blade has no significant influence on forming adventitious bud.
