王百合雄性不育突变体小孢子败育的细胞学研究

摘    要:对王百合雄性不育突变体‘白天使’小孢子败育的过程进行了显微观察 .结果显示 :①花粉母细胞减数分裂异常 .花粉母细胞早期出现质壁分离及多核仁现象 ;减数分裂末期Ⅰ ,部分花粉母细胞畸形 ,核结构破坏 ;减数分裂过程中 70 %花粉母细胞出现了染色体结构或数量的变异 .②绒毡层行为异常 .花粉母细胞形成时期至减数分裂末期I ,绒毡层细胞液泡化 ,细胞质稀少 ;有的过度膨胀 ,彼此挤压 ,细胞粘连成团块或分隔成数块 ;单核花粉粒时期 ,绒毡层细胞质浓厚 ,细胞间间隙小 ,与中层细胞联系紧密 ,没有解体 .推测减数分裂及绒毡层行为的异常共同导致了小孢子的败育 .

Abstract:Observation on microspore abortion of ‘White angel was made by means of light microscopy. ①The meiosis of PMC was showed to be abnormal. At its early stage, PMC cytoplasm cell wall separation and multi micronucleoli were observed; At meiosis tetophase I, some PMC were showed to be irregular and their nucleus structure destroyed; Chromosome structural or quantitative aberrance was observed in 70% PMC during meiosis. ②The tapetum action was revealed to be abnormal. From the early PMC stage to meiosis telophase I, large vacuole and sparse cytoplasm in PMC was observed. Some PMC were showed to expand excessively, extrude each other, conglutinate to gobbet or separate to small parts. At the stage of single nucleus pollen, tapetum cells were showed to possess dense cytoplasm, join with its conjoint cell with little space, connect with middle cell closely and not disaggregate. It was proposed that the meiosis and the tapetum action abnormality caused the microspore abortion.
