| 摘 要： | 本试验以番茄"中蔬四号"品种为试材,对不同激素组合、不同种类外植体、子叶不同部位及接种方式对番茄不定芽再生的影响进行了研究,同时还筛选了种子消毒的最佳方法和不定芽生根的最佳培养基。研究结果表明,番茄种子表面消毒宜采用70%乙醇浸泡30s,然后再用20%的次氯酸钠加土温消毒20min;子叶中间段为最佳外植体,茎段次之,真叶的诱芽率最低;子叶诱芽的最佳培养基为MS+ZT2mg/L+IAA0.1mg/L,且背面朝上放置更有利于不定芽的分化;不定芽生根的最佳培养基为1/2MS+IAA0.3mg/L。这项研究为番茄转化体系的建立奠定了基础。
