枯草芽孢杆菌HAS中几丁质酶基因的克隆分析

摘    要:为揭示枯草芽孢杆菌菌株HAS对甘蔗黑穗病菌抑制作用的机理,选取对多种病原真菌有抑制作用的几丁质酶进行克隆与分析。结果表明:菌株HAS基因组中含有1个ORF全长834bp的几丁质酶编码序列,其具有编码278个氨基酸的几丁质酶蛋白,该序列同GenBank上登录的几丁质酶蛋白(Bacillus subtilis subsp.subtilis str.168)(ref|NP_390566.1)的同源性最高,达98%。推测该几丁质酶在菌株HAS抑制甘蔗黑穗病菌过程中可能起关键作用。

Abstract:The chitinase with an inhibition effect on various pathogenic fungi was cloned and analyzed to reveal the inhibition mechanism of B.subtilis HAS against sugarcane S.scitaminea.Results: The genome of HAS strain contains a chitinase coding sequence with 834 bp and the chitinase coding sequence has chitinase protein with 278 animo acids.The homology between the chitinase coding sequence and B.subtilis subsp.subtilis str.168)(ref| NP_390566.1)listed in GenBank is up to 98%.In conclusion, the chitinase in B.subtilis HAS strain may play a vital role to inhibit sugarcane S.scitaminea.
