烟叶烟碱含量与腐胺N-甲基转移酶基因表达间的相关性

摘    要:为探明烟碱合成与积累机制,采用大田试验方法,测定云烟87和红花大金元在四川省6个产区烤后烟叶的烟碱含量,检测其在旺长期、现蕾期及成熟期PMT基因的表达量,并分析PMT基因表达量与烟碱含量之间的相关性。结果表明:四川不同产区不同烤烟品种的烟碱含量差异较大,云烟87和红花大金元在古蔺烟区烟碱含量最高,分别为3.40%和4.37%;云烟87在米易烟区烟碱含量最低,为1.27%,红花大金元在会理烟区烟碱含量最低,为1.36%。经相关性分析,2个品种的烟碱含量均与成熟期PMT基因表达量呈极显著正相关,与现蕾期基因表达量呈显著(红花大金元)或极显著(云烟87)正相关。PMT基因表达量是影响烟碱含量的决定因素之一。

Abstract:In order to explore the relationship between tobacco nicotine content and gene expression of the putrescine N-methyl-transferase (PMT), six tobacco growing areas in Sichuan Province were selected for the determination of nicotine content of flue-cured tobacco of Yun 87 and Honghuadajinyuan, and their PMT gene expression in different growth periods such as vigorous stage, budding stage and mature stage were measured.Correlation analysis between nicotine content and gene expression levels was conducted.Results: Nicotine content of flue-cured tobacco varied across tobacco cultivation areas in Sichuan Province.Among all the six tobacco cultivation areas, nicotine contents of Yun 87 and Honghuadajinyuan in Gulin were 3.40% and 4.37%, respectively, and were the highest.While that of Yun 87 in Miyi(1.27%) and Honghuadajinyuan(1.36%) were the lowest.Correlation relationship analysis showed that the nicotine content of the two varieties was significantly positively correlated with the expression of PMT gene in mature stage, and in bud stage, this correlation is positively extremely significant for Yun 87 and positively significant for Honghuadajinyuan.The results indicate that PMT gene expression is one of the determinants of nicotine content in tobacco leaves.
