| 摘 要： | 根据猪繁殖与呼吸综合征病毒(PRRSV)美洲型标准株(ATCC VR-2332)的部分ORF6及ORF7保守序列和已发表的猪圆环病毒株PCV-2(DQ195679)的ORF-2基因保守序列,设计合成了2对特异引物,分别扩增出大小为426 bp和631 bp特异性基因片段,通过优化RT-PCR和PCR条件,最终建立了可同时检测PRRSV和PCV-2的复合PCR诊断方法。应用此方法分别对河南省不同地区送检的15头份病、死猪的血清、淋巴结、肺、肝等组织进行检测,结果8头份PRRSV阳性,5头份PCV-2阳性,其中3头份PCV-2和PRRSV同时为阳性,其余猪为阴性,健康猪对照样品全部阴性。结果表明,PRRSV和PCV-2复合PCR诊断方法具有高度特异性和敏感性,可用于兽医临床诊断。

| Abstract: | Based on conserved sequences in ORF6 and ORF7 fragments of PRRSV American strain(ATCC VR-2332) and that in ORF2 of PCV-2(DQ195679)strain,two pairs of specific primers were respectively designed,through optimizing of RTPCR and PCR conditions,multiplex PCR for detection of PRRSV and PCV-2 was established.Fifteen organ samples from different farms of Henan province were respectively detected by the duplex PCR,out of which eight were PRRSV positive,five were PCV-2 positive,three were both PRRSV and PCV-2 positive,the control samples were negative,suggesting that the multiplex PCR of PRRSV and PCV-2 was a rapid, specific and sensitive diagnostic method. |
