杜长大猪CIDEa基因克隆及其组织表达分析

摘    要:本实验旨在克隆杜长大猪(三元杂交猪)诱导细胞凋亡DNA片段化因子45样效应子A(CIDEa)基因序列,通过生物学软件分析CIDEa基因序列,利用实时荧光定量(qPCR)方法检测杜长大猪不同组织中CIDEa基因mRNA的表达量。实验成功获得杜长大猪CIDEa基因CDS区,长度为660 bp,共编码218个氨基酸。杜长大猪CIDEa基因与NCBI已公布野猪、牛、马、犬、人和家鼠的CIDEa基因同源性分别为99.2%、84.8%、81.5%、80.6%、80%、76.1%。与野猪CIDEa基因相比,发生5处碱基突变,分别为第99 C-T、第355 T-C、第609 G-A、第627 C-T,为同义突变,第173 T-C脯氨酸突变为亮氨酸。CIDEa蛋白二级结构预测结果显示,α-螺旋占45.41%,无规则卷曲占33.49%,延伸链占15.14%,β-转角占5.96%;CIDEa蛋白三级结构与二级结构相一致。qPCR结果显示,CIDEa基因在腹脂和皮下脂肪中表达量最高,在肝脏、肾脏、肺脏和心脏中均有表达,在背最长肌中表达量最低。结论表明:CIDEa基因在脂肪沉积中可能起着重要作用。

Abstract:The purpose of this experiment was to clone the cell death-inducing DNA fragmentation factor 45-like effector A(CIDEa) gene sequence in DLY pigs(ternary hybrid pigs), analyze the CIDEa gene sequence through biological software, and detect the expression of CIDEa gene mRNA in different tissues of DLY pigs by real-time fluorescence quantitative(qPCR).The CDS region of CIDEa gene of DLY pigs was successfully obtained, with a length of 660 bp and encoding 218 amino acids. The homology of CIDEa gene of DLY pigs and that of Sus scrofa, Bos taurus, Equus caballus, Canis lupus familiaris,Homo sapiens and Mus musculus published by NCBI were 99.2%, 84.8%, 81.5%, 80.6%, 80% and 76.1% respectively.Compared with Sus scrofa CIDEa gene, there were 5 base mutations: 99 th C-T, 355 th T-C, 609 th G-A and 627 th C-T were synonymous mutations, and 173 T-C proline was leucine. The secondary structure prediction results of CIDEa protein show that, α-helix accounted for 45.41%, irregular curl accounted for 33.49%, and extended chain accounted for 15.14%, β-corner accounts for 5.96%. The tertiary structure of CIDEa protein was consistent with the secondary structure. The expression of CIDEa gene was the highest in abdominal fat and subcutaneous fat, expressed in liver, kidney, lung and heart, and the lowest in longissimus dorsi muscle. Conclusion: CIDEa gene may play an important role in fat deposition, and may participate in the regulation of fat deposition.
