摘    要:影响水稻花培一次成苗的主要因素为:(1)基本培养基、激素配比和基因型对一次成苗有重要作用.基本培养基以N′6的为最好;激素配比以2,4—D0.02+NAA2.50+KT4-6mg/L为佳;3种基因型一次培养的绿苗率大小顺序为粳型品种C57>籼粳交(早香17×C57)杂种F1>籼型品种早香17.(2)培养基保持pH值5.8、蔗糖浓度8.5%,并添加丝氨酸、脯氨酸、精氨酸、天冬氨酸、丙氨酸各40mg/L和谷氨酰胺400mg/L,所获一次培养的绿苗率最高;添加0.1%活性炭的有抑制作用.(3)低温(9-10℃)预处理对一次成苗有重要作用.早香17以预处理5-10d的为宜,C57以10—16d的较好.恒温处理优于变温处理的.高温(30℃)预培养的作用不大.接种后进行暗培养对一次成苗有利.

Abstract:The main factors affecting the one-step seedling formation were as follows: (1) The one-step seedling formation was influenced by following three factors, that is, basal media, different hormone combinations and genotypes. In the experiment of basal medium N'6, the highest green plantlet frequencies were obtained. The green plantlet frequncies had been siginificantly increased in the medium adding 2,4-D 0.02+NAA 2.50+KT 4-6 mg/ L. For the three genotypes, experiments showed different levels of green plantlet frequencies, with the highest level in japonica C57, the middle level in indica/japonica hybrid (Zaoxiang 17/C57) and the lowest level in indica Zaoxiang 17. (2) It was revealed in the experiment of one-step culture that the highest green plantlet frequencies could be obtained when pH value was kept with 5.8 and sucrose concentration with 8.5% in the medium adding 40 mg /L of serine, proline, arginine, aspartic acid and alanine, respectively, and 400 mg / L of glutamine. 0.1 % active charcoal, added in media, would inhibit seedling generation. (3) Low temperature (9-10) ) pre-treatment played an important role in one-step seedling formation. For Zaoxiang 17, suitable treatment period lasted for 5-10 days, but for C57, 10-16 days were suitable treating length. Meanwhile, constant temperature treatment showed better affect on one-step seedling formation than varied temperature treatment did. however, high temperature treatment (30℃) revealed little effect on one-step seedling formation. Culture of inoculated anther in dark appeared to be a favorable practice to increase green plantlet frequencies.
