雀麦花叶病毒德国株系与英国株系病毒粒子的比较研究

摘    要: 采用多种方法对雀麦花叶病毒德国株系(BMV-G)及英国株系(BMV-E)粒子的提纯及RNAs的提取效果进行了比较,结果分别以聚乙二醇(PEG)沉淀法及两相酚提取法为佳。聚丙烯酰胺凝胶(PAA)电泳证明BMV-G具有5个RNA组份,比BMV-E (代表典型株系)多一个RNA-3a,其分子量为0.52×106,介于RNA-3及RNA-4之间。另外变性及不变性RNAs分子量的测定结果也基本相同。薄层电泳分析指出BMV二种株系RNAs的碱基比也基本一致,同于Paul及Huth的报道。经34% CsCl平衡等密度离心得知BMV-G及BMV-E均具有4种粒子组份,其中最重的粒子的浮力比重值分别为1.3513及1.3534,而最轻的则分别达1.2993及1.3045。

Abstract:Different methods of virus purification and RNAs extraction are made with brome mosaic virus German strain(BMV-G) and England strain(BMV-E).It is proved that the polyethylene glycol(PEG) purified method and two phases phenol extracted method are best for virus purification and RNAs extraction respectively.The results of polyacrylamide gel electrophoresis indicate that comparing with BMV-E(represent typical strain),the BMV-G has five RNA components and including an additional component-RNA-3a(mol.wt.-0.52×106) between RNA-3 and RNA-4.The estimated molecular weights of nature and denature RNAs are similar.By means of cellulose thin layer electrophoresis analysis,the basic ratios of BMV-G and BMV-E are same and similar to Paul and Huth's report also.According to the 34% CsCl equilibrated density centrifuge experiments,BMV-G and BMV-E can all form four bands in the tubes.The heaviest virus particle's buoyant density values are 1.3513and 1.3534 g/cm3 respectively,and the lightest are 1.2993 and 1.3045.
